Ceramide Kinase (FLAG Tag), active human recombinant protein

Human ceramide kinase (CERK) (GenBank Accession No. NM_022766), 1-537(end) with C-terminal Flag tag, MW= 60 kDa, expressed in baculovirus expression system. A unit of ceramide kinase activity is defined as the amount of enzyme required to produce 1 pmole of ceramide-1-phosphate/min at 37°C. Assay condition: 20 mM HEPES pH7.2, 80 mM KCl, 3 mM CaCl2, 10 mM MgCl2, and 2 mM DTT, 2.5 mM Cardiolipin, and 0.5 mM diethylenetriamine pentaacetic acid in H2O), OG/CL/DTPA solution (3.75 % beta-Octylglucoside, Ceramide (1.25 mM in 5% Triton X-100 containing PBS),500 uM ATP and at 37°C for 25 min. ATP reduction is detected using Kinase-Glo(R) Luminescent Kinase Assay Platform (Promega, Madison, WI).Application: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
