Plasmide

Plasmide sind kleine, zirkuläre, doppelsträngige DNA-Moleküle meist bakteriellen Ursprungs. Ein Plasmid liegt in der Zelle oftmals episomal vor, das heißt es repliziert unabhängig vom Genom der Zelle. Dies führt in der Regel zu einer hohen Zahl an Kopien des Plasmids. Plasmide werden primär als Klonierungsvektoren zur Vervielfältigung und Expression rekombinanter DNA verwendet. Darüber hinaus werden Plasmide auch als Expressionsvektoren zur biotechnologischen Produktion von Proteinen in Zellkulturen genutzt. Zu Forschungszwecken ist hierfür eine große Anzahl rekombinanter Plasmide erhältlich, die cDNAs verschiedenster Gene tragen. Ferner wurden Plasmide sowie Backbones entwickelt, die für verschiedene Expressionssysteme und Spezies optimiert sind.

Plasmide
