Biologische Puffer

Puffersubstanzen ermöglichen es, den pH-Wert einer Reaktionslösung zu kontrollieren, indem sie Hydronium- oder Hydroxidionen abfangen. Der pH-Wert bleibt durch den Puffer nach Zugabe von Säuren oder Basen weitgehend konstant. Dies ist besonders in biochemischen Reaktionen wichtig, da die Ladung von biologischen Molekülen wie Proteinen und Nukleinsäuren stark vom pH-Wert abhängig ist. Folglich sind biologische Lösungen wie Blut oder Speichel durch Phosphat- und/oder Carbonat-Ionen gepuffert. Diese Funktion wird in der molekularbiologischen und biochemischen Forschung, z.B. im Medium von Zellkulturen, meist durch sogenannte „Good-Puffer“ oder PBS ersetzt.
