In linkage disequilibrium mapping of the MHC region in 80 British parent-offspring trios, Wei and Hemmings(2000) found that NOTCH4 was highly associated with schizophrenia. Repression of Notch4 resolved ataxia and reversed the disease progression, demonstrating that it is not only sufficient to induce but also required to sustain the disease. Protein function: Functions as a receptor for membrane-bound ligands Jagged1, Jagged2 and Delta1 to regulate cell-fate determination. Upon ligand activation through the released notch intracellular domain (NICD) it forms a transcriptional activator complex with RBPJ/RBPSUH and activates genes of the enhancer of split locus.
