Anti-Glypican 1 (Glypiated Proteoglycan 1)

Glypican 1 (glypiated proteoglycan 1) is a 200kD, highly glycosylated member of the GPI-linked Glypican family of proteins. It is expressed by both epithelial and mesodermally-derived cells, and appears to use its proteoglycan adduct to regulate growth factor signaling and availability. Human Glypican 1 is synthesized as a 535 amino acid (aa) proprecursor that contains a 507 mature region (aa 24-530) and a 28 aa C-terminal prosegment. There are three potential splice variants. One shows an alternate start site at Met73, a second incorporates the same start site with a 65 aa substitution for aa 295-558, and a third shows a deletion of aa 184-558. Mature human Glypican 1 is 91% aa identical to mouse Glypican 1. Applications: Suitable for use in Direct ELISA, Immunocytochemistry, Flow Cytometry and Western Blot. Other applications not tested. Recommended Dilutions: Flow Cytometry: 2.5ug/10e6 cells using MDA-MB-231human breast cancer cell line , Western Blot: 0.1ug/ml, Immunocytochemistry: 5-15ug/ml using immersion fixed MDA-MB-231 human breast cancer cell line, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
