Rusfertide

Rusfertide is a peptide mimetic of hepcidin, a hormone that binds to the iron exporter ferroportin to reduce systemic iron levels. It induces internalization of ferroportin in a cell-based assay (EC50 = 6.12 nM). Rusfertide (1 mg/kg) increases the survival of red blood cells (RBCs) in an Hbbth3/+ mouse model of beta-thalassemia. It reduces transferrin saturation (TSAT) levels by approximately 60% following a single dose at 2.5 mg/kg in a mouse model of hemochromatosis using mice lacking homeostatic iron regulator (HFE). Rusfertide prevents hepatic iron deposition in the same model when administered at a dose of 2.5 mg/kg every other day for two weeks..
