Osteopontin is a single-chain polypeptide with a molecular weight of ~35kD. It is a glycoprotein first identified in osteoblasts. It is biosynthesized by a variety of tissue types including preosteoblasts, osteoblasts, osteocytes, extraosseous cells in the inner ear, brain, kidney, deciduum, placenta, odontoblasts, some bone marrow cells, hypertrophic chondrocytes, macrophages, smooth muscle, and endothelial cells. Osteopontin is overexpressed in a variety of cancers, including lung cancer, breast cancer, colorectal cancer, stomach cancer, ovarian cancer, melanoma and mesothelioma. It may contribute to kidney stone formation and both glomerulonephritis and tubulointerstitial nephritis and is also found in atheromatous plaques within arteries. Research has implicated osteopontin in excessive scar-forming and a gel has been developed to inhibit its effect. Applications: Suitable for use in ELISA and Western Blot. Other applications have not been tested. Recommended Dilutions: ELISA: 1:64,000, Western Blot: 1-3ug/ml. An ~30kD band observed in lysates of cell lines Jurkat and MOLT4. This band corresponds to the cleavage product from the 60kD glycoprotein (calculated MW of 33.8kD according to NP_000573.1). , Optimal dilutions to be determined by the researcher. Peptide Blocking: Corresponding peptide is available for Peptide Blocking studies. See O8064-07-P. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
