Anti-Melanoma, gp100 (pMel17, Melanocyte Protein PMEL, Melanocytes Lineage-specific Antigen GP100)

By immunohistochemistry, 138569-AF specifically recognizes a protein in melanocytes and melanomas. 138569-AF reacts with junctional and blue nevus cells and variably with fetal and neonatal melanocytes. Intradermal nevi, normal adult melanocytes, and non-melanocytic cells are negative. It does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin. 138569-AF labels formalin-fixed, paraffin-embedded melanomas and other tumors showing melanocytic differentiation. Applications: Suitable for use in ELISA, Flow Cytometry, Immunofluorescence and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 0.5-1ug/10e6 cells, Immunofluorescence: 1-2ug/ml, Immunohistochemistry (FFPE): 1-2ug/ml/30min at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 minutes followed by cooling at RT for 20 minutes. , Optimal dilutions to be determined by the researcher. Positive Control: SK-MEL-28 cells or Melanoma, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
