Cytosolic malic enzyme (ME1) catalyzes the conversion of malate and NADP+ to pyruvate and NADPH (1,2). NADPH is then used for fatty acid biosynthesis and lipogenesis (1,2). Cytosolic malic enzyme was shown to mediate high fat diet-induced adiposity (1). Mitochondrial malic enzyme (ME2) preferentially uses NAD+ to catalyze the conversion of malate to pyruvate with the concomitant generation of NADH (2). Recent studies have demonstrated that the tumor suppressor p53 regulates cell metabolism and proliferation by repressing the expression of both cytosolic malic enzyme and mitochondrial malic enzyme (3). Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1:1000. Incubate membrane with diluted antibody in 5% w/v BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight., Immunoprecipitation: 1:50, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
