Lysosome-associated membrane protein-1(LAMP1), also known as CD107a, is a 100-130kD member of the LAMP family of glycoproteins. It is expressed in lysosomal and plasma membranes of macrophages, NK and T-cells, and with LAMP2, is essential for the formation of phagolysosomes. On the cell surface, it also presents carbohydrates to selectins. Mature human LAMP1 is a 389aa type I transmembrane glycoprotein. It contains a 354aa luminal/extracellular domain (ECD) (aa28-381) and a 12aa cytoplasmic tail (aa405-416). The ECD has two large looping regions (aa28-193 and 227-381) plus multiple N-and O-linked glycosylation sites. There is one potential splice variant that shows a 26aa substitution in the signal sequence. Over (aa28-380), human LAMP1 shares 64% aa identity with mouse LAMP 1. Applications: Suitable for use in Flow Cytometry, Western Blot, Immunocytochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 25ug/ml, Western Blot: 2ug/ml, Immunocytochemistry: 10ug/ml, Optimal dilutions to be determined by the researcher. Hybridoma: Myeloma cells with B cells from Balb/c mice. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
