PF-06446846 is an inhibitor of proprotein convertase subtilisin kexin type 9 (PCSK9) translation. It inhibits translation of mRNA encoding PCSK9 by isolated human ribosomes when used at a concentration of 100 µM. PF-06446846 also inhibits PCSK9 secretion by Huh7 cells (IC50 = 0. µM). In vivo, PF-06446846 (50 mg/kg) decreases plasma levels of Pcsk9, total cholesterol, and LDL cholesterol in rats.
