CF3-2N6-UC18 is an ionizable cationic and chloroquine-derived lipid that has been used in the generation of lipid nanoparticles (LNPs) for the delivery of mRNA in vitro and in vivo. LNPs containing CF3-2N6-UC18 and encapsulating a Cy5-mRNA reporter exhibit increased cellular uptake via micropinocytosis and improved endosomal escape compared with LNPs containing non-chloroquine-derived lipids. LNPs containing CF3-2N6-UC18 selectively accumulate in mouse lymph nodes after intravenous administration.
