Thawing and Culturing Cells Confirm that the vial remains deeply frozen upon delivery, as cells are shipped on dry ice to maintain optimal temperatures during transit. Upon receipt, either store the cryovial immediately at temperatures below -150°C to ensure the preservation of cellular integrity, or proceed to step 3 if immediate culturing is required. For immediate culturing, swiftly thaw the vial by immersing it in a 37°C water bath with clean water and an antimicrobial agent, agitating gently for 40-60 seconds until a small ice clump remains. Perform all subsequent steps under sterile conditions in a flow hood, disinfecting the cryovial with 70% ethanol before opening. Carefully open the disinfected vial and transfer the cell suspension into a 15 ml centrifuge tube containing 8 ml of room-temperature culture medium, mixing gently. Centrifuge the mixture at 300 x g for 3 minutes to separate the cells and carefully discard the supernatant containing residual freezing medium. Gently resuspend the cell pellet in 10 ml of fresh culture medium. Cytogenetically, OV-90 cells carry mutations in tumor suppressor genes and oncogenes frequently implicated in ovarian cancer, including TP53 and BRCA2, as well as alterations in TGF-beta receptor type II and CDKN2A. These mutations reflect the genomic instability commonly observed in high-grade serous ovarian carcinomas. Gene expression profiling of OV-90 reveals a distinct molecular signature consistent with its tumor origin. Comparative microarray analyses have shown that OV-90's transcriptomic profile diverges significantly from that of normal ovarian surface epithelium, with strong upregulation of genes involved in proliferation, DNA damage response, and invasion. Moreover, among the ovarian cancer lines studied, OV-90 clusters with other aggressive tumor-derived lines rather than with those derived from indolent disease, making it a useful model for investigating high-risk disease biology. Its expression patterns also align with clinical markers of poor prognosis, further supporting its utility in preclinical research focused on aggressive ovarian cancer subtypes. In systems biology and pharmacogenomic studies, OV-90 has been included in large-scale transcriptomic and proteomic analyses, including the Cancer Cell Line Encyclopedia (CCLE) and proteomic atlases.
