Mouse INHbB (Inhibin Beta B) ELISA Kit

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Mouse INHbB. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Mouse INHbB. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Mouse INHbB, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Mouse INHbB in the samples is then determined by comparing the OD of the samples to the standard curve. Protein Function: Inhibins and activins inhibit and activate, respectively, the secretion of follitropin by the pituitary gland. Inhibins/activins are involved in regulating a number of diverse functions such as hypothalamic and pituitary hormone secretion, gonadal hormone secretion, germ cell development and maturation, erythroid differentiation, insulin secretion, nerve cell survival, embryonic axial development or bone growth, depending on their subunit composition. Inhibins appear to oppose the functions of activins , Activin B is a dimer of alpha and beta-B that plays a role in several essential biological processes including embryonic development, stem cell maintenance and differentiation, haematopoiesis, cell proliferation and wound healing (PubMed:8125256). Signals through type I receptor ACVR1C, abundantly expressed in pancreatic beta cells, and type II receptors like ACVR2A. Upon ligand binding, these receptors phosphorylate intracellular signaling mediators SMAD2 and SMAD3, which form a complex with SMAD4, translocate to the nucleus, and regulate gene expression. Plays a crucial role in the induction of hepcidin by inflammation through activation of ACVR1C and subsequent phosphorylation of SMAD1/5/8 (PubMed:18480258). Regulates adipocyte lipid metabolism by decreasing non-esterified fatty acids and glycerol release and increases intracellular triglyceride content (PubMed:20382119). Stimulates wound healing by promoting cell migration and hair follicle regeneration through the JNK and ERK signaling pathways downstream of RHOA (PubMed:28396222, PubMed:35690801) , Inhibin B is a dimer of alpha and beta-B that plays a crucial role in the regulation of the reproductive system by inhibiting the secretion of follicle-stimulating hormone (FSH) from the anterior pituitary gland. Thereby, maintains reproductive homeostasis in both males and females. Acts as a more potent suppressor of FSH release than inhibin A (By similarity).
