Human IL-1beta ELISA Kit

Interleukin-1beta (IL-1beta) is a potent stimulator of bone resorption whose gene is mapped to 2q14 and has been implicated in the pathogenesis of high bone turnover and osteoporosis. IL-1beta, a prominent microglia-derived cytokine, causes oligodendrocyte death in co-culture with astrocytes and microglia, but not in pure culture of oligodendrocytes alone. It also can cause nuclear export of a specific NCOR co-repressor complex, resulting in de-repression of a specific subset of nuclear factor-kappa-B-regulated genes. Furthermore, microenvironmental IL-1beta and, to a lesser extent, IL-1a, are required for in vivo angiogenesis and invasiveness of different tumor cells. Additionally, the cooperation of IL-1beta and PDGFB induces contractile-to-synthetic phenotype modulation of human aortic smooth muscle cells in culture. Moreover, the association with disease may be explained by the biologic properties of IL-1beta, which is an important proinflammatory cytokine and a powerful inhibitor of gastric acid secretion. Human IL1-beta-specific monoclonal antibodies are pre-coated onto 8 x 12 divisible strips. The human specific detection monoclonal antibodies are biotinylated. The test samples and biotinylated detection antibodies are subsequently added to the wells and then washed with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The intensity of yellow is proportional to the amount of human IL-1beta captured in strips. The human IL-1beta detected in sera, plasma, body fluids, tissue lysates or cell culture supernatants with this ELISA kit is mainly activated mature protein.
