Human BMP5 (Bone Morphogenetic Protein 5) Microsample ELISA Kit

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human BMP5. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human BMP5. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human BMP5, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human BMP5 in the samples is then determined by comparing the OD of the samples to the standard curve.Detection Range: 15.63-1000 pg/mL. Sensitivity: 5.2 pg/mL. Initiates the canonical BMP signaling cascade by associating with type I receptor BMPR1A and type II receptor BMPR2 (PubMed:11580864). In turn, BMPR1A propagates signal by phosphorylating SMAD1/5/8 that travel to the nucleus and act as activators and repressors of transcription of target genes (PubMed:11580864, PubMed:29321139). Can also signal through non-canonical pathway such as MAPK p38 signaling cascade to promote chondrogenic differentiation (PubMed:20402566).
