Human Immunoglobulin G2 (IgG2) is one of the four primary subclasses of IgG, which is the most abundant protein in human serum. IgG2 accounts for approximately 10-20% of total plasma protein. Compared to other IgG subclasses, IgG2 exhibits distinctive structural characteristics such as having a more rigid hinge region due to a higher number of inter-heavy chain disulfide bridges, specifically up to four additional ones. This rigidity limits the flexibility of the IgG2 molecule. Additionally, IgG2 lacks certain glycan residues present in the hinge region of IgG1, resulting in differences in effector functions like antigen binding, immune complex formation, complement activation, and interaction with Fc receptors.IgG2 plays a crucial role in the immune response, particularly in protection against bacterial infections involving capsules. However, individuals who exhibit a deficiency in IgG2 may experience increased susceptibility to diseases caused by encapsulated bacteria. Protein function: Constant region of immunoglobulin heavy chains. Immunoglobulins, also known as antibodies, are membrane-bound or secreted glycoproteins produced by B lymphocytes. In the recognition phase of humoral immunity, the membrane-bound immunoglobulins serve as receptors which, upon binding of a specific antigen, trigger the clonal expansion and differentiation of B lymphocytes into immunoglobulins- secreting plasma cells. Secreted immunoglobulins mediate the effector phase of humoral immunity, which results in the elimination of bound antigens (PubMed:22158414, PubMed:20176268). The antigen binding site is formed by the variable domain of one heavy chain, together with that of its associated light chain. Thus, each immunoglobulin has two antigen binding sites with remarkable affinity for a particular antigen. The variable domains are assembled by a process called V-(D)-J rearrangement and can then be subjected to somatic hypermutations which, after exposure to antigen and selection, allow affinity maturation for a particular antigen (PubMed:17576170, PubMed:20176268).
