Product Description: WNT inhibitory factor 1 (WIF1) is a secreted protein composed of 379 amino acids, featuring an N-terminal signal sequence, a unique and conserved 150-amino-acid WIF domain, five epidermal growth factor (EGF)-like repeats, and a hydrophilic C-terminal tail. WIF1 functions as a lipid-binding protein that directly interacts with a broad range of canonical and non-canonical Wnt ligands, such as Wnt3a, Wnt4, Wnt5a, Wnt7a, Wnt9a, and Wnt11, thereby inhibiting their ability to bind to membrane-bound receptors and suppressing Wnt/beta-catenin signaling. Structural studies have confirmed its capacity to bind additional extracellular proteins, including olfactomedin 1 and connective tissue growth factor, suggesting a multifaceted regulatory role in extracellular signaling. In disease, WIF1 acts as a tumor suppressor, with its expression frequently downregulated in cancers such as osteosarcoma due to promoter hypermethylation, restoration of WIF1 expression in deficient tumor cells significantly reduces tumor growth and metastasis in vivo.
