Product Description: Proline-rich AKT1 substrate 1 (AKT1S1), also known as PRAS40, is a critical regulatory protein that interacts with the AKT signaling pathway, which is essential for cell growth, survival, and metabolism. AKT1S1 functions as a negative regulator of AKT by binding to its phosphorylated form, thereby inhibiting its activity. This protein is characterized by its proline-rich domain, which facilitates interactions with various signaling molecules. Dysregulation of AKT1S1 has been implicated in several diseases, particularly cancer and metabolic disorders. For instance, decreased expression of AKT1S1 has been associated with increased tumorigenesis and poor prognosis in cancers such as breast and prostate cancer, where enhanced AKT activity promotes cell proliferation and survival. Additionally, alterations in AKT1S1 expression have been linked to insulin resistance and type 2 diabetes, highlighting its role in metabolic regulation. Protein Function: Negative regulator of the mechanistic target of rapamycin complex 1 (mTORC1), an evolutionarily conserved central nutrient sensor that stimulates anabolic reactions and macromolecule biosynthesis to promote cellular biomass generation and growth (PubMed:17277771, PubMed:17386266, PubMed:17510057, PubMed:29236692). In absence of insulin and nutrients, AKT1S1 associates with the mTORC1 complex and directly inhibits mTORC1 activity by blocking the MTOR substrate-recruitment site (PubMed:29236692).
