This technology has immense possibilities to evolve beyond infectious disease to reach targets once considered undruggable and treat a near infinite number of conditions. With LNPs, it is now possible to move beyond traditional small molecules into nucleic acid-based therapies. With the right design, it should be possible to engineer LNPs to suit any number of payloads for vaccination, protein replacement therapy, and therapeutic gene knockdown. Cayman Chemical: Oxidative Stress Guide

Oxidative stress occurs when there is an imbalance between the production of chemically reactive species such as reactive oxygen, nitrogen, and sulfur species (ROS, RNS, RSS) and antioxidant defense mechanisms. Uncontrolled oxidation can disrupt redox signaling and cause injury to cellular components like lipids, proteins, and nucleic acids, resulting in irreparable damage and eventual cell death. Oxidative stress can be evaluated directly by measuring reactive species or indirectly by the associated damage to lipids, proteins, and nucleic acids. Although direct measurement of these reactive species is desirable, indirect methods are often more reliable due to the relative stability of damage markers on biomolecules compared to the transient nature of reactive species. Multiple biomarkers of oxidative damage have been identified for different macromolecules (e.g., proteins, lipids, and DNA/RNA). The success of mRNA-based COVID-19 vaccines could not have been possible without decades of research on lipid-based drug delivery (LBDD). LBDD systems are highly versatile and have been used to deliver various bioactive molecules to targeted cells and tissues. LBDD has several advantages over conventional drug delivery methods, including increased drug stability, bioavailability, and distribution. Lipid nanoparticles (LNPs) are a significant advancement for the delivery of oligonucleotide-based therapeutics.
