Protein Construction: Recombinant Human Toll-Interacting Protein is produced by our E.coli expression system and the target gene encoding Ala2-Pro274 is expressed with a 6His tag at the C-terminus. Sequence: Ala2-Pro274. Fusion tag: C-6His Endotoxin: < 1.0 EU per µg as determined by the LAL method. Apparent Molecular Mass: 31 kDa. Protein function: Toll-Interacting Protein (TOLLIP) is a member of the tollip family. TOLLIP localizes to the cytoplasm. It contains one C2 domain and one CUE domain. TOLLIP is an inhibitory adaptor protein for Toll-like receptors (TLR). The Toll-like receptors pathway is a part of the immune system that recognize structurally conserved molecular patterns of microbial pathogens, resulting in an inflammatory immune response. TOLLIP constitutes a complex with Tom1 to regulate endosomal transferring of ubiquitinated proteins. TOLLIP can negative regulate Toll-like receptors signaling, which may limit the production of proinflammatory mediators during the process of inflammation and infection. Protein function: Component of the signaling pathway of IL-1 and Toll-like receptors. Inhibits cell activation by microbial products. Recruits IRAK1 to the IL-1 receptor complex. Inhibits IRAK1 phosphorylation and kinase activity (PubMed:11751856). Connects the ubiquitin pathway to autophagy by functioning as a ubiquitin- ATG8 family adapter and thus mediating autophagic clearance of ubiquitin conjugates. The TOLLIP-dependent selective autophagy pathway plays an important role in clearance of cytotoxic polyQ proteins aggregates (PubMed:25042851). [The UniProt Consortium]
