MTAP (Methylthioadenosine phosphorylase) is important in polyamine metabolism and for the salvage of both adenine and methionine. It is involved in the metabolism of MeSAdo (methylthioadenosine), a by-product of the synthesis of the polyamines spermidine and spermine. MTAP is deficient in many cancers because the MTAP gene is co-deleted with the tumor suppressor p16. Source: Recombinant human PKM2, full length, with N-terminal His tag, expressed in E. coli. Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. , Enzyme Activity: , A 50ul PKM2 reaction is conducted in a buffer containing 50mM Tris, pH 7.4, 10mM MgCl2, 50mM KCl, 2mM ADP, 10mM phosphoenolpyruvate (PEP) at RT for 15 minutes. ATP production is detected using Kinase-Glo(R) Luminescent Kinase Assay Platform. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 6 months.
