UGQuencher-2 NHS ester, which is equivalent to BHQ 2 NHS ester, is a dark quencher featuring a polyaromatic-azo structure that prevents any intrinsic fluorescence. This non-emissive quencher is widely used in qPCR probes and other fluorescence-quenched probe applications. Its broad and strong absorption range (560-670 nm) makes it highly effective as an acceptor in FRET applications, particularly when paired with orange to far-red emitting dyes such as Cy3, Cy5, ROX, Texas Red, Alexa Fluor(R) 546, 555, 594, 633, 647, and IRDye(R) 650. The NHS ester (or succinimidyl ester) group is a commonly used amine-reactive functionality, enabling covalent conjugation to primary amines in proteins (e.g., lysine residues), amine-modified oligonucleotides, and other amine-containing molecules. Labeling is most efficient under mildly alkaline conditions (pH 7-9), resulting in the formation of stable amide bonds. This combination of properties makes UGQuencher-2 NHS ester a preferred choice for developing fluorescence-quenched probes and biomolecular conjugates.
