RhoGAP1 is a 439aa intracellular GTPase activator for the Rho proteins (Rho, Rac and Cdc42). It has a proline-rich domain, Src homology 3 (SH3) binding domain and three consensus box regions characteristic of RhoGAP. It stabilizes charges developing on the guanine nucleotide during the transition state for GTP hydrolysis and thus contributes to GAP-stimulated catalysis and converts the Rho proteins to their putatively inactive GDP-bound state with Cdc42 being the preferred substrate. Along with Rho proteins they modulate cytoskeletal changes and cell signaling. It is known to regulate the basal apoptosis of cells through the JNK pathway and also play a crucial role in regulating Rho-mediated signaling pathways. Recent reports suggest that this protein may also have a positive input to cell proliferation. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.5-2ug/ml, Optimal dilutions to be determined by the researcher. Positive Control: HeLa, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
