NOD2 is a member of the apoptosis regulating protein family that includes caspase recruitment-domains, as well as Apaf-1 and NOD1. It contains two N-terminal CARDs, a nucleotide binding domain (NBD), and multiple C-terminal leucine-rich repeats (LRRs). NOD2 is expressed in monocytes (whereas NOD1 is expressed in multiple tissues). NOD2 plays a role in regulating NF-kappaB, acts as an intracellular receptor for bacterial lipopolysaccharides, and contributes to inflammatory bowel disease (IBD) and Crohn's disease. Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunofluorescence/Immunocytochemistry, and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:100-1:2000. Tested on NOD2 transfected 293T lysates and HT29 human colorectal adenocarcinoma lysates. Endogenous detection has some background. Immunofluorescence/ Immunocytochemistry: 1:10-1:500, Immunohistochemistry (Paraffin): 1:10-1:500, Immunoprecipitation: 1:10-1:500, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
