The sodium/calcium exchanger of cardiac sarcolemma rapidly transports calcium during excitation-contraction coupling and is the dominant myocardial calcium efflux mechanism. The sodium/calcium exchanger uses the transmembrane sodium gradient to catalyze countertransport of calcium against its electrochemical gradient in a 3 sodium : 1 calcium exchange. Sodium/calcium exchange activity is present in excitable cells and in non-excitable cells. Applications: Suitable for use in Western Blot, ELISA, Immunohistochemistry, Immunofluorescence/Immunocytochemistry, Flow Cytometry and Immunoprecipitation. Other applications not tested. Recommended Dilutions: ELISA: 1:100-1:2000, Flow Cytometry: 1:100, Immunofluorescence (IC): 1:200, Immunohistochemistry: 1:20-1:50, Immunohistochemistry (Paraffin): 1:100, Immunoprecipitation: 1:10-1:500, Western Blot: 1:100-1:2000. Detects a 120kD protein representing the sodium/calcium exchanger from guinea pig cardiac extract. The bands seen at 70kD and 160kD represent a proteolytic fragment and non-reduced exchanger respectively. T-tubular membrane., Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
