Anti-Myogenin (myf4), clone 4H208

Myogenin1 is a basic helix loop helix (bHLH) transcription factor that is specifically expressed in myogenic cells. It is therefore a useful marker for tumors of the muscle lineage2, being strongly expressed in alveolar rhabdomyosarcomas. Gene knockout studies in mice have shown that myogenin is essential for muscle maturation3. Applications: Can be used for detection of myogenin (myf4) expression in myogenic cells by Immunocytochemistry(4), Immunohistochemistry, Immunofluorescence and by western blotting(4). Whole cell lysate from the human Rh30 alveolar rhabdomyosarcoma cell line can be used as positive control and a band of approximately 34kD band should be detected. The antibody can also be used for immunoprecipitation and gel shift assays. Recommended Dilution: Immunocytochemistry(4)/Immunofluorescence: 5ug/ml in 1% BSA in PBS, Immunohistochemistry: Frozen 5-10ug/ml, Immunohistochemistry: Paraffin 10-20ug/ml, Western Blot(4): 2ug/ml in 5% non-fat milk buffer. Western blot analysis of Myogenin expression in various small-round cell tumor lines using anti-Myogenin mAb. The antibody strongly reacted with a band of appox. 34kD in alveolar rhobdomyosarcoma cell line (Rh30). All other small-round cell tumor lines were negative. PFSK-1 (a primitive neuroectodermal tumor cell line), EB2 (a lymphoma cell line), SK-N-SH ( a neuroblastoma cell line), SJSA-1 (Ewing's sarcoma cell line). Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
