ERK2 is a protein serine/threonine kinase that is a member of the extracellular signal-regulated kinases (ERKs) which are activated in response to numerous growth factors and cytokines. Activation of ERK2 requires both tyrosine and threonine phosphorylation that is mediated by MEK. ERK2 is ubiquitously distributed in tissues with the highest expression in heart, brain and spinal cord. Activated ERK2 translocates into the nucleus where it phosphorylates various transcription factors (e.g., Elk-1, c-Myc, c-Jun, c-Fos, and C/EBP beta). Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommneded Dilution: Western Blot: 0.5-2ug/ml detected p42MAP Kinase (erk2) in human A431 carcinoma cells, mouse 3T3 fibroblasts and rat L6 skeletal fibroblasts. , Immunoprecipitation: 5ug of this antibody immunoprecipitated p42MAPK (erk2) from 3T3 cells as confirmed by Western Blot using the monoclonal anti-erk2., Optimal dilutions to be determined by the researcher. Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo, I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo, Positive Control Included: F4209: Fibroblast Cell, 3T3/A31, Add 2.5ul 2-mercaptoethanol/100ul of lysate. Boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for Western Blot analysis. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
