FOXP3 (a 381aa protein) is a member of the forkhead/winged-helix family of transcriptional regulators and is highly conserved across mammals. FOXP3 is essential for normal immune homeostasis. FOXP3 is stably and constitutively expressed at a high level in CD25 + CD4 positive regulatory T cells, at a low level in CD4-positive/CD25-negative cells, and is absent in CD4-negative/CD8-positive T cells. FOXP3 may be a master regulatory gene and a more specific marker of regulatory T cells than other T cells. Applications: Suitable for use in Flow Cytometry, Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: Band is seen at ~48kD in FOXP3 transfected and CD25/CD4 positive regulatory T cell lysates., Optimal dilutions to be determined by the researcher. Positive Control: Mouse CD4+CD25+ T cells, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
