CXCR3 is a G protein­ coupled chemokine receptor that binds the alpha chemokines MIG (CXCL9), IP­10 (CXCL10), and I­TAC (CXCL11). CXCR3 is expressed on activated T cells, B cells, and NK cells. Applications: Suitable for use in Neutralization, Flow Cytometry and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Neutralization (ND50): ~0.3-1.5ug/ml in the presence of 7ng/ml of recombinant human CXCL11/l-TAC, using the human CXCR3 transfected BaF/3 cells in a chemotaxis assay. , The exact concentration required to neutralize human cell surface CXCR3 activity is dependent on the ligand concentration as well as the number of CXCR3 receptors present on the cell surface. , Flow Cytometry: 0.25ug/10e6 cells using whole human blood lymphocytes and HEK293 human cell line transfected with human CXCR3 and eGFP., Immunohistochemistry: 8-25ug/ml detected CXCR3 in immersion fixed paraffin-embedded sections of human tonsil. , Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-06F: IgG, H&L (FITC) (X-Adsorbed) Pab Gt xMo I1904-23: IgG, X-Adsorbed (FITC) Pab Gt xMo, Hybridoma: Mouse myeloma with B cells obtained from a mouse inoculated with human CXCR3 transfected NS0 mouse myeloma cells. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
