AMICA (adhesion molecule, interacting with CXADR antigen 1), also known as JAML, is a 65kD, heavily glycosylated transmembrane protein that belongs to the junctional adhesion molecule (JAM) subset of the immunoglobulin superfamily. JAM family molecules contribute to intercellular connections within epithelial and endothelial cell layers, and mediate their interactions with various hemopoietic cells. The human AMICA cDNA encodes a 384aa precursor that includes a 19aa signal sequence, a 256aa extracellular domain (ECD) with two Ig­ like domains, a 21aa transmembrane segment, and a 98aa cytoplasmic domain. Alternative splicing may generate isoforms with N­ and C­ terminal deletions. In contrast to other JAM family proteins, AMICA does not contain a cytoplasmic PDZ­ binding motif. Applications: Suitable for use in Flow Cytometry, ELISA and Western Blot. Other applications not tested. Recommended Dilutions: Flow Cytometry: 2.5/10e6 using Human peripheral blood monocytes., ELISA (Capture): 0.2-0.8ug/ml can be used as a capture reagent in a human AMICA sandwich immunoassay in combination with the biotinylated human AMICA detection antibody (A1372-31H5) and recombinant human AMICA as the standard. Shows <0.3% crossreactivity with recombinant mouse AMICA and recombinant human JAM-A, JAM-B and JAM-C., Western Blot: 0.1ug/ml using Recombinant Human AMICA/JAML Fc Chimera , Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
