Lectins are derived from the extracts of plants, animals, viruses and microorganisms. They are known to agglutinate red blood cells. These agglutinins can select cell types according to blood group activities utilizing sugar-binding mechanisms. Lectins form precipitates with glycoconjugates and are useful for identifying or separating oligosaccharides with identical sugar compositions such as galactose, mannose or glucose. PHA consists of two molecular species, an erythroagglutinin (PHA-E) which has low mitogenic activity and high erythroagglutinin activity, and leucoagglutinin (PHA-L) which has high mitogenic and leucoagglutinating activity, but very low erythroagglutinating activity. PHA-E is not blood group specific, but agglutination can be inhibited by certain oligosaccharides. PHA-P is the protein form of PHA prior to separation and purification of erythroagglutinin and leucoagglutinin. PHA-M is the mucoprotein form. , P. vulgaris lectin consists of 4-subunit isoforms: E4, consisting of 4 E subunits, L4, consisting of 4 L subunits, E3L1, E2L2, and E1L3. Source: Phaseolus vulgaris-P, Red Kidney Bean, Purity: ~99% by SDS PAGE. Purified by affinity chromatography. May contain traces of salt. Appearance: Supplied as a white, lyophilized powder. Reconstitute with assay buffer. Subunit Molecular Weight: 26-30kD, Carbohydrate Specificity: Recognizes a complex branched chain oligosaccharide structure, Inhibitory Carbohydrate: Not inhibited by simple sugars. Activity: < 5mg/ml will agglutinate type O human erythrocytes. < 0.5mg/ml will agglutinate neuraminidase treated erythrocytes. < 5ug/ml will agglutinate rabbit erythrocytes. Solubility: Soluble in most liquids, Quality Control: SDS-PAGE, Agglutination assay. Storage and Stability: Lyophilized powder is stable for 12 months at -20°C. Reconstituted product must be stored in aliquots at -20°C.
